He studied theology, under Mezzofanti at Bologna, and in 1824 became professor of oriental languages at Pisa University.

He had the opportunity, in Florence, to meet Champollion. A deep and true friendship grew up between them, nourished by mutual esteem and a common interest in the study of hieroglyphics.
It was Rosellini who taught the first official course in Europe, at the University of Pisa, on the history, language and antiquities of Egypt.
